磷酸鹽輔助合成的紅色熒光碳點(diǎn)用于光動力學(xué)治療 徐雅蘭,賀依民,王嬋*,宋啟軍* 江南大學(xué) 化學(xué)與材料工程學(xué)院,無錫,214122 *Email: wangchan@jiangnan.edu.cn ;qsong@jiangnan.edu.cn 光動力學(xué)治療(PDT)是利用光敏劑吸收的能量傳遞給的氧氣,以生成活性強(qiáng)的單線態(tài)氧(1O2),并將其用于癌癥診療的一種手段,具有實(shí)施方便、創(chuàng)傷小、選擇性好等特點(diǎn)。然而,傳統(tǒng)光敏劑光穩(wěn)定性較差、1O2產(chǎn)率較低,其中吩噻嗪類光敏劑(如亞甲基藍(lán))易與DNA的磷酸基團(tuán)結(jié)合,產(chǎn)生致癌副作用。本課題組從開發(fā)新型光敏劑、提高1O2產(chǎn)率出發(fā),以亞甲基藍(lán)為碳源,磷酸鹽輔助水熱合成了紅色熒光碳點(diǎn)(R-CDs),所得R-CDs的水溶性優(yōu)異、光穩(wěn)定性好、1O2產(chǎn)率高(0.75),同時由于磷酸基團(tuán)的引入,克服了DNA被侵染的缺點(diǎn)。 Phosphate-assisted synthesis of red fluorescent carbon dots for photodynamic therapy Yalan Xu, Yimin He, Chan Wang*, Qijun Song* School of Chemical&Material Engineering, Jiangnan University, Wuxi, 214122 A new kind of red emissive carbon dots (R-CDs) was prepared hydrothermally by using methylene blue (MB) as the sole carbon source in KH2PO4 solution. The resultant R-CDs exhibit excellent water solubility, photostability and high singlet oxygen (1O2) yield (0.75), thus they have been successfully applied to the PDT study in vitro. More importantly, the R-CDs show non-infective property to DNA, which is substantially different to their precursor MB. The structure of R-CDs was comprehensively characterized both experimentally as well as by density functional theory (DFT) calculation. This study not only provides a rational strategy for preparation of highly efficient PDT material, but also gives an insight into the mechanism of 1O2 generation.
